Differences in regeneration capacity between five elite varieties of maize (Zea mays L.) and the DNA differences between embryogenic callus and nonembryogenic callus

Cytokinin KT and 6-BA were evaluated for their function in callus induction and plant regeneration. The results showed that cytokinins improved the regeneration capacity of maize and 6-BA was more effective than KT. Five maize (Zea mays L.) varieties of commercial importance were screened for the regeneration capacity of calluses formed from their immature embryo. The difference in the rate of regeneration between 78% of Putian No. 1 and 8% of Yedan No. 9 were pronounced. The mode of plant regeneration from type I callus also varied with the genotypes. Suyu No. 1 tended to regenerate via somatic embryogenesis, while Putian No. 1 was likely to regenerate via organogenesis. Maize callus lost the capacity of embryogenesis during long-time subculture. The results of randomly amplified polymorphic DNA (RAPD) showed that its genome DNA was modified in tissue culture.
